A diploma, masters and Ph.D. position is available in the Molecular Physics department at the Fritz Haber Institute of the Max Planck Society (FHI) in Berlin, Germany. The project is focussed on the analysis of protein glycosylation using the promising technique of ion mobility spectrometry.
Glycosylation is one of the most abundant types of post-translational modifications found in proteins. The involved oligosaccharides are termed glycans and usually exhibit a branched and stereochemically complex structure. Currently, glycans are typically characterized using mass spectrometry-based techniques (MS). Measuring the molecular weight of a sugar, however, immediately poses a fundamental problem: entire classes of the constituting monosaccharide building blocks exhibit an identical atomic composition and, consequently, also an identical mass. Therefore, carbohydrate MS data can be highly ambiguous and often it is simply not possible to clearly assign a particular molecular structure. Within this project a novel, interdisciplinary approach to characterize complex sugars by incorporating ion mobility spectrometry as an additional gas-phase separation dimension should be developed and applied. Experimentally, this involves a variety of biochemical and bioanalytical techniques such as the enzymatic cleavage of the sugar form the investigated gycoprotein, the instrumental analysis using a commercially available, hybrid ion mobility mass spectrometer as well as the analysis of complex datasets using in-house developed software tools.
The applicant should have a background in biochemistry, (bio)organic chemistry and/or analytical chemistry. Basic experience with chromatographic separation techniques as well as mass spectrometry-based techniques would be helpful, but is not required.

The Fritz-Haber Institute (Molekülphysik) offers a Ph.D. and Posdoctoral position(s).
A Ph.D. and/or postdoctoral position is available in the molecular physics department at the Fritz-Haber-Institut der Max-Planck-Gesellschaft (FHI) in Berlin, Germany. The research involves spectroscopic studies of mass/charge selected (bio-)molecules using the FHI infrared free electron laser. The data is used to gain fundamental insight into the structure, and function of biological molecules and their aggregates. In one experiment, those (bio-)molecular ions are incorporated in ultracold liquid helium droplets, which allows for measuring IR spectra at high resolution (see for example Phys.Chem.Chem.Phys., 17, 21902 (2015)). In a different experiment, in addition to mass/charge selection, conformer selection is performed using ion mobility methods. This allows for the recording of IR spectra of species that have the same mass/charge ratio, but differ in conformation, protonation site or aggregation state (see for example J. Am. Chem. Soc., 137, 4236 (2015)).
- degree in physical chemistry, experimental physics or related fields
- experience in laser spectroscopy on gas phase molecules, gas-phase molecular beam methods and/or mass-spectrometry (required for the post-doctoral and desired for the graduate student position)
